OK, now that you have your layers set up, you will want to paint in your base colors. This is where you will be establishing the basic color layout. Keep everything fairly dark/neutral, since you will be adding your brightest, most saturated details and highlights towards the end. Your base color should support the shadows. I generally like to have some bit of a complimentary color in the shadows/shaded areas. That's why the bas background colors tend towards cool gray-blues and greens, because I am already thinking the creature will be in the warmer red/yellow range.
Step 09:
More color exploration. Starting to hone in on the colors for this guy. I am still working in the layer underneath the drawing. Starting to work in some colors for my background.

Step 10:
Now I am beginning to work on the top layer, starting in the area that I think will be the brightest, most saturated part of the piece: the head. Build your color slowly. I use a flat round brush at about 30 percent opacity to build the color up. In the case of this piece, I am using a pretty small brush too, which will end up giving be a cool texture as a slowly build the color using small scritchy lines. In subsequent steps, I continue to build the color in the rest of the creature.
